Dutch vote could affect how EU tackles debt crisis

The free-market Liberal party and the center-left Labor party, running neck in neck in Dutch polls, could end up forming a centrist coalition and leave extremist parties out of government.

Dutch voters on Wednesday were picking a new parliament in a test of support for stringent austerity measures which are set to influence the way the European Union tackles the debt crisis.

The export-dependent Netherlands is a founding member of the EU and has long been a staunch supporter of the bloc's open market. But many Dutch voters have begun questioning their role in the now 27-nation EU since the debt crisis erupted in 2009, feeling that their wealthy nation is paying too high a price to help bail out countries like Greece and Portugal.

Even so, the two parties now leading the polls remain committed to Europe, though they differ on how to tackle its crisis.

"We are all in the same boat. There is no way we can turn our back on the EU," said Lodewijk van Groeningen at a polling station close to parliament, before he sped off on his bike.

As the Dutch voted, European Commission President Manuel Barroso was appealing for greater EU unity.

"We cannot continue trying to solve European problems just with national solutions," he said in his annual State of the Union address to Parliament in Strasbourg, France.

"A deep and genuine economic and monetary union ... means ultimately that the present European Union must evolve," he added. "And let's not be afraid of the words. We will need to move toward a federation of nation states."

Meanwhile, domestic spending cuts are increasingly unpopular as the Dutch economy has barely recovered from a recession last year.

Wednesday's election has boiled down to a tight race between the free-market Liberal (VVD) party of Prime Minister Mark Rutte and the center-left Labor Party led by Diederik Samsom, with smaller parties trailing.

The Dutch proportional representation system guarantees a coalition government and whichever party wins the most in the 150-seat House of Representatives will take the lead in choosing the parties that make up the next ruling coalition.

Despite their differences on Europe, Mr. Rutte and Mr. Samsom could win enough seats to wind up in the same coalition government, together with one more centrist party.

While critical of a strict austerity-only solution to the debt crisis, Labor backed Rutte at crucial moments to approve bailout funds and endorse European-level solutions to prevent the debt crisis from spinning out of control.

"The latest polls suggest that three parties could govern without having to depend on any extremist party," said Piotr Kaczynski of the Center for European Policy Studies.

Rutte is a close ally of German Chancellor Angela Merkel and supports her austerity agenda for reining in the debt crisis. Samsom is closer to French President Fran?ois Hollande, who favors government spending to spur economic growth.

Libyan officials: US ambassador killed in attack

TRIPOLI, Libya (AP) ? Libyan officials say the U.S. ambassador and three other Americans have been killed in an attack on the U.S. consulate in the eastern city of Benghazi by protesters angry over a film that ridiculed Islam's Prophet Muhammad.

The officials say Ambassador Chris Stevens was killed Tuesday night when he and a group of embassy employees went to the consulate to try to evacuate staff. The protesters were firing gunshots and rocket propelled grenades.

All of the officials ? three in all ? hold senior security positions in Benghazi.

They are deputy interior minister for eastern Libya Wanis al-Sharaf; Benghazi security chief Abdel-Basit Haroun; and Benghazi city council and security official Ahmed Bousinia.

Millions of DNA switches that power human genome's operating system are discovered

ScienceDaily (Sep. 5, 2012) ? The locations of millions of DNA 'switches' that dictate how, when, and where in the body different genes turn on and off have been identified by a research team led by the University of Washington in Seattle. Genes make up only 2 percent of the human genome and were easy to spot, but the on/off switches controlling those genes were encrypted within the remaining 98 percent of the genome.

Without these switches, called regulatory DNA, genes are inert. Researchers around the world have been focused on identifying regulatory DNA to understand how the genome works. Using a new technology developed with funding from the National Human Genome Research Institute's ENCODE (ENCyclopedia Of DNA Elements) project, UW researchers created the first detailed maps of where regulatory DNA is located within hundreds of different kinds of living cells. They also compiled a dictionary of the instructions written within regulatory DNA -- the genome's programming language.

The findings are reported in two papers appearing in the Sept. 5 online issue of Nature.

"These breakthrough studies provide the first extensive maps of the DNA switches that control human genes," said Dr. John A. Stamatoyannopoulos, associate professor of genome sciences and medicine at the University of Washington, and senior author on both papers. "This information is vital to understanding how the body makes different kinds of cells, and how normal gene circuitry gets rewired in disease. We are now able to read the living human genome at an unprecedented level of detail, and to begin to make sense of the complex instruction set that ultimately influences a wide range of human biology."

Here are the key results:

1) The first detailed maps of regulatory DNA switches that make up the genome's 'operating system'.

The instructions within regulatory DNA are inscribed in small DNA 'words' that function as the docking sites for special proteins involved in gene control. In many cases, these switches are located far away from the genes that they control. To map the regulatory DNA regions, the researchers harnessed a special molecular probe -- an enzyme called DNaseI -- that snips the genome's DNA backbone. Under the right conditions, these snips occur precisely where proteins are docked at regulatory DNA. By treating cells with DNase I and analyzing the patterns of snipped DNA sequences using massively parallel sequencing technology and powerful computers, the researchers were able to create comprehensive maps of all the regulatory DNA in hundreds of different cell and tissue types. They found that of the 2.89 million regulatory DNA regions they mapped, only a small fraction -- around 200,000 -- were active in any given cell type. This fraction is almost totally unique to each type of cell and becomes a sort of molecular bar code of the cell's identity. The researchers also developed a method for linking regulatory DNA to the genes it controls. The results of these analyses show that the regulatory 'program' of most genes is made up of more than a dozen switches. Together, these findings greatly expand the understanding of how genes are controlled and how that control may differ between normal and diseased cells.

2) The first extensive map of regulatory protein docking sites on the human genome reveals the dictionary of DNA words comprise the genome's programming language.

The instructions for turning genes on and off are written in DNA switches called regulatory DNA. These switches are scattered throughout the non-gene regions of the human genome. Having mapped the locations of the regulatory DNA switches, UW researchers wanted to know what made them tick. These regions contain small chains of DNA 'words' that make up docking sites for special regulatory proteins involved in gene control. The human genome contains hundreds of genes that make such proteins.

However, current technologies only allow such proteins to be studied one at a time. They also lack the accuracy to resolve the DNA letters to which the proteins dock. As a result, most of the actual DNA words recognized by regulatory proteins in living cells were unknown. To find them, the researchers employed a simple, powerful trick that enabled them to study all the proteins at once.

Instead of trying to see proteins directly, they looked for their shadows or 'footprints' on the DNA. To accomplish this, they again turned to the DNaseI enzyme that snips the DNA backbone within regulatory DNA. Prior work had shown that DNaseI likes to snip DNA next to regulatory protein docking sites, but not within the docking site itself. By using next-generation DNA sequencing technology, the researchers analyzed hundreds of millions of DNA backbone breaks made when cells were treated with DNaseI. They then used a powerful computer to resolve millions of protein footprints. In total, they identified 8.4 million such footprints along the genome, some of which were detected in many cell types. Next, they compiled all of the short DNA sequences to which the proteins were docked. They analyzed them using a software algorithm that required hundreds of microprocessors working simultaneously. This revealed that more than 90 percent of the protein docking sites were actually slight variants of 683 different DNA words -- essentially a dictionary of the genome's programming language.

"These findings significantly advance the understanding of how the instructions for controlling genes are written and organized throughout the genome, and how combinations of different instruction sets function together to control genes, often at great distance along the genome," Stamatoyannopoulos said. "The broad spectrum of cell and tissue types included in these analyses provide an incredibly rich resource that can be mined immediately by researchers around the world to illuminate how the genes they are studying are controlled."

The scientists determined that genes are connected in a complex web. In this web, regulatory DNA regions typically control one or at most a few genes, but genes receive inputs from large numbers of regulatory regions. The researchers also found evidence for a combinatorial code that helps match regulatory DNA with the right genes. Another key finding was that the regulatory DNA controlling genes involved in cancer and other types of 'immortal' cells that can keep on growing indefinitely appears to acquire mutations at a different rate than other kinds of regulatory DNA. This result points to a previously unknown link between genome function and patterns of DNA variation in individual human genomes. The finding may have implications for understanding susceptibility to cancer.

The findings reported in these papers are expanded upon in two related papers to be published simultaneously in the journals Science and Cell. In the Science paper, UW researchers further expanded the regulatory DNA maps, and compared them with genetic maps of human disease. Their studies revealed that most DNA variants associated with specific human diseases or clinical traits are located in regulatory DNA rather than in gene sequences. In the Cell paper, the researchers describe using the detailed information on regulatory protein docking sites to create a comprehensive map of how those proteins are wired.

Biodiversity Found to Increase During Warm Periods in Earth's History

A new approach contradicts previous research that reported an inverse correlation between high temperatures and biodiversity

Rather than kicking off the expected cycles of extinction, periods of warming in Earth's history were accompanied by increased biodiversity, according to a report published this week. But this does not mean that the mass extinctions that are taking place today, with Earth warming at an unprecedented rate, will be reversed in future.

Researchers examined the number of known families of marine invertebrates, as well as sea-surface temperatures, over the course of 540 million years of Earth's history1. They found that when temperatures were high, so was biodiversity. When temperatures fell, biodiversity also declined.

The results contradict previous work, including findings from lead author Peter Mayhew's group2, that reported an inverse correlation between high temperatures and biodiversity.

The reason for the about-face, says Mayhew, an evolutionary ecologist at the University of York, UK, is that the earlier work measured fossil diversity by tallying the first and last appearances of each group of species, then assuming that the creatures existed only during the intervening years. This might sound logical, but overlooks the fact that some geological periods are better studied than others.

To correct this, the new study looked only at the well-sampled periods. And, instead of interpolating organisms' presence from origination and extinction dates, it merely tallied species groups present during each period.

Even so, given that climate change is generally viewed as disruptive, Mayhew admits it was a "big surprise" to find that eras of warming were accompanied by increases in biodiversity. The work also provided a solution to another puzzle, Mayhew says. Tropical ecosystems are known to be Earth's most diverse, and the tropics would be expected to expand during warm eras. Yet in the past these eras were thought to be species-poor compared with cooler ones. The new results resolve that contradiction.

Speed kills
Warming produces both extinctions and originations, and in the past the originations of new species have outstripped the loss of old ones, says Mayhew. But this does not mean that today's climate change will be beneficial.

"The rate of change is very important," Mayhew says. For diversity to rise, he explains, new species need to evolve. And that takes between thousands and millions of years ? much slower than the rate at which extinctions are likely to occur with today's rapid change.

Scott Wing, a palaeobiologist at the Smithsonian Institution in Washington DC, agrees. "This article has nothing to say about the effects of global warming at any timescale of interest to most humans," he says. But he adds, "this is definitely of interest to evolutionary biologists, palaeontologists and ecologists seeking to understand very broad patterns of diversity".

However, Shanan Peters, a palaeobiologist at the University of Wisconsin-Madison, isn't so sure the paper is noteworthy. Its primary result, he points out, is to overturn Mayhew's own prior finding and bring the long-term diversity results into line with ecological common sense. "Palaeobiologists and climatologists have long referred to warm intervals as 'climate optima'," he notes, "precisely because it is during such times that palm trees and alligators inhabit the Arctic and life appears to be diverse and flourishing."

More interesting, he says, will be the next generation of results, when palaeobiologists turn their attention to major climate transitions, such as that which may be occurring now.

Sleep in style: Airline giving away $4,000 eye masks

The excitement of finding a crystal-adorned eye mask in an amenity kit may preclude sleep.

Travelers hoping to avoid jet lag on a long-haul flight often don black eye masks in hopes of getting some shut-eye.?

They may look bland, but they work.

But on Thursday, five lucky Virgin Atlantic passengers will be able to snooze in considerably better style wearing a limited-edition mask, decorated with $4,000 worth of Swarovski crystals.


The airline introduced new amenity kits for its economy, premium economy and first-class passengers on Sept. 1. To celebrate, it's tucking five swanky Swarovski eye masks ? each with more than 3,000 red, white and blue crystals???in the amenity kits handed out in the economy section of five flights. The crystals were all hand applied (10 hours per mask) by Saima Anwar, an artist who also creates crystal eyelashes for celebrities such as Katy Perry.

Thursday?s winners could include passengers on any of the airline?s flights to or from 11 cities in North America, including New York, Chicago, Boston and Los Angeles.

Five eyeshades have been created to mark the launch of new amenity kits which are now appearing on board all Virgin Atlantic flights.

?As the majority of our flights leave at night, arriving in London in the morning, we want to make sure our guests are well rested to begin their days, whether they be traveling for business or pleasure,? said Chris Rossi, senior vice president North America.

Virgin Atlantic has been sweet on Swarovski for a while. Since 2003, the airline?s first-class cabins have featured Swarovski crystals on the cabin walls. Curtains adorned with more than 1,000 Swarovski crystals were recently added to the revamped first-class section on the airline?s A330 aircraft.

While only five passengers will score the cool, crystal-encrusted eye-masks, the new, complimentary amenity kits are sure to be keepers. Each economy class kit contains one of the airline?s signature red eyeshades decorated with one of six sunglass designs, including one with heart-shaped frame and another in a shutter style.

The premium economy kits are charcoal gray pouches made from recycled plastic bottles. The pouches have silk linings are designed to double as travel wallets.

Virgin Atlantic?s first-class passengers will be issued amenity kits made from the same recycled material, but their pouches will be big enough to be reused as holders for tablet devices or e-readers.

Why upgrade an amenity that many other airlines don?t even offer to travelers in the coach cabin? ?To enhance the customer experience to travelers in any class by offering a number of on- and off-board amenities not found with other carriers,? said Rossi. ?

Or, as the airline states in a news release, because it?s ?all part of Virgin Atlantic?s commitment to sustainability, and going green (and gorgeous!).?

Sept. 3 (Bloomberg) -- German Finance Minister Wolfgang Schaeuble warned against placing too much faith in the European Central Bank?s bond-buying plans as pressure grows on Spain and Italy to decide whether to seek assistance to lower borrowing costs.

Whatever proposals ECB President Mario Draghi announces on Sept. 6 must fall within the Frankfurt-based central bank?s mandate, Schaeuble said in an interview today on Deutschlandfunk radio. Germany won?t accept ECB financing of state budgets, he said.

?We have to be very careful that we don?t raise false expectations,? Schaeuble told the broadcaster in Berlin. ?It has to remain very clear, state debt can?t be financed through monetary policy. Therefore we can?t have a decision -- we would think it very wrong -- that?s not covered by the ECB mandate.?

European leaders are awaiting the details of the ECB?s plan and a German high-court decision next week that will determine whether policy makers have the bailout funding required to tackle the euro crisis almost three years after it emerged in Greece. The stakes for those countries at the heart of the contagion were underscored last week when Spain?s government said it will inject emergency funding into Bankia group, its third-largest lender, which posted a first-half loss.

Spanish Prime Minister Mariano Rajoy reiterated over the weekend that his government will wait to see what the ECB unveils before seeking assistance to address rising bond yields. Draghi said Aug. 2 that any ECB action would be conducted in tandem with the region?s rescue funds and depend on governments agreeing to strict conditions.

Spain?s 10-year yield slid 3 basis points to 6.80 percent as of 10:32 a.m. in Madrid after climbing 44 basis points last week. The euro was little changed at $1.2572.

?Draghi?s announcement of intervention shows the robust will of the ECB to solve the problem,? Rajoy told Spain?s ABC, Germany?s Bild am Sonntag, France?s Le Journal du Dimanche and Italy?s Corriere della Sera in a joint interview published over the weekend. ?I will await the results of the ECB and then make a decision that?s good for Spain and for the euro.?

Finance ministers are also struggling to agree on a plan to help integrate the banking sector of the 17-member currency union by placing the bloc?s bank supervision in the hands of the ECB. Schaeuble took issue with European Commissioner Michel Barnier, the EU?s financial services chief who said last week that he wanted the supervision to be in effect from next year.

Schaeuble ?Surprised?

?I think that?s a bit unrealistic,? Schaeuble told Deutschlandfunk, saying that he was ?surprised? as euro group ministers hadn?t yet discussed the draft proposal. ?It?s always this way -- there?s an interview and the media jumps on it and then expectations are raised that can?t be met,? he said.

The so-called banking union is a condition for enabling the euro bailout funds to finance banks directly, offering the prospect of breaking the link between ailing credit sectors and the sovereign-debt market that?s driving the crisis.

Bailout funding will be on hold until Germany?s Federal Constitutional Court decides on Sept. 12 whether to suspend the euro-area?s permanent rescue fund, the European Stability Mechanism. Only with its endorsement can the legislation be approved and the 500 billion-euro ($628 billion) ESM go into operation.

The ECB?s bond plan would also be held up without bailout money, since any scheme would involve central-bank intervention working alongside joint funding to ease yields. The ECB?s Executive Board will send a list of options to the 17 governors tomorrow, a day before the Governing Council convenes in Frankfurt, three central bank officials said Aug. 31.

Options under consideration include sovereign-bond yield caps and targets on spreads over German bunds, the officials said. One said there has also been discussion about making purchases in a range of asset classes and not just government bonds. An ECB spokesman declined to comment.

Red Cross chief will request Assad's help with Syria's humanitarian crisis

The Red Cross head is traveling to Syria to request that President Bashar al-Assad make it easier for humanitarian workers to reach civilians facing deteriorating living conditions.

Peter Maurer?said he would also continue efforts to gain access for his agency to?Syria's detention centers ? which rights groups say hold tens of thousands of people rounded up during the 17-month-old conflict, including teenagers.

"At a time when more and more civilians are being exposed to extreme violence, it is of the utmost importance that we and the Syrian?Arab Red Crescent?succeed in significantly scaling up our humanitarian response," he said in a statement.

"An adequate humanitarian response is required to keep pace with needs, which have been growing exponentially," added Maurer, who took over as president of the independent organization from?Jakob Kellenberger?on July 1.

Obama: GOP ideas better suited for the last century

"You might as well have watched it on a black and white TV," Obama told a cheering crowd of thousands in Urbandale, outside Des Moines. The president was in Iowa ? a pivotal battleground state ? on the first stop of a push that will take him to the Democratic National Convention in Charlotte, N.C.

Obama recalled how his upset victory over Hillary Clinton in the Iowa caucuses four years ago had revived his campaign at a time when pundits were predicting his political demise. "And it will be you, Iowa, who will choose the path we take from here."

The president took several shots at the Republican convention in Tampa, noting Romney did not mention Afghanistan in his acceptance speech on Thursday, and accusing his political foes of having outdated ideas. "It was something to behold. Despite all the challenges that we face in this new century what they offered over those three days was, more often than not, an agenda that was better suited for the last century," he said. "It was a re-run. We'd seen it before."

"If you didn't DVR it, let me recap it for you: Everything's bad, it's Obama's fault, and Governor Romney is the only one who knows the secret to creating jobs and growing the economy," he said. "That was the pitch."

"And when Governor Romney had his chance to let you in on his secret, he did not offer a single new idea. Just retreads of the same old policies that have been sticking it to the middle class for years," Obama charged.

A spokesman for Mitt Romney's campaign, Ryan Williams, hit back.

"President Obama dubbed this trip the 'Road to Charlotte' but his policies have taken us on a road to declining incomes, higher unemployment and more uncertainty for the middle class. And in the face of a record of failure, he offered no new solutions, just misleading attacks," Williams said.

Obama's greatest vulnerability is the sour economy, still sputtering nearly four years after he took office vowing to fix it. Stubbornly high unemployment and sluggish growth weigh down his hopes for a second term. The president's response has been to argue that he inherited an economic crisis of historic dimensions and that Romney's approach is to favor the rich at the expense of the middle class.

Republican vice presidential hopeful Paul Ryan explicitly took on that argument explicitly in his speech. "We are four years into this presidency. The issue is not the economy as Barack Obama inherited it, not the economy as he envisions it, but this economy as we are living it," he said.
